Protective Role of IL-17 in Pulmonary Tuberculosis Relapse

چکیده
Abstract Tuberculosis (TB) recurrence following drug treatment and apparent clinical cure affects an estimated 700,000 people annually is increasingly associated with relapse of initial Mycobacterium tuberculosis (Mtb) infection. Cellular immunity by T helper (Th) cells, including Th17 populations important during active TB. The contribution other Th to immune pressure Mtb clearance TB chemotherapy poorly understood. We used Mtb-HN878 for a paucibacillary model infection in WT, IL17RA-KO, second study using WT conditional Th17-KO (CD4-Cre-RORγt flox/flox) mice identify the cellular sources protective role IL-17 therapy. Assessment disrupted lung multi-parameter flow cytometry demonstrated that expand TB, retract after therapy, subsequently display moderate re-expansion relapse. Flow cytometric analysis also revealed expansion contraction +cells among both non-lymphocyte CD3 −lymphocyte active, post-drug, phase Compared IL-17RA-KO had greater mycobacterial burden These results demonstrate critical cells signaling protection against regrowth chemotherapy. Further, it suggests lymphoid non-lymphoid innate system contribute IL-17-mediated NHI grant R01AI147948

Unexpected Role for IL-17 in Protective Immunity against Hypervirulent Mycobacterium tuberculosis HN878 Infection
Mycobacterium tuberculosis (Mtb), the causative agent of tuberculosis (TB), infects one third of the world's population. Among these infections, clinical isolates belonging to the W-Beijing appear to be emerging, representing about 50% of Mtb isolates in East Asia, and about 13% of all Mtb isolates worldwide. In animal models, infection with W-Beijing strain, Mtb HN878, is considered "hyperviru...
متن کاملThe critical role of epithelial-derived Act1 in IL-17- and IL-25-mediated pulmonary inflammation.
IL-25 initiates, promotes, and augments Th2 immune responses. In this study, we report that Act1, a key component in IL-17-mediated signaling, is an essential signaling molecule for IL-25 signaling. Although Act1-deficient mice showed reduced expression of KC (CXCL1) and neutrophil recruitment to the airway compared with wild-type mice in response to IL-17 stimulation, Act1 deficiency abolished...

OBJECTIVE To identify patients at high risk of relapse after anti-tuberculosis (TB) therapy or with poor long-term outcomes. METHODS Fifty-one patients with pulmonary TB: 7 were classified as high association with both cavitations on initial chest radiography and positive sputum smear/cultures after two months of anti-TB treatment (HA group); 19 medium association (MA, one risk alone); and 25...
متن کاملA STUDY OF MYCOBACTERIUM TUBERCULOSIS DRUG RESISTANCE IN PULMONARY TUBERCULOSIS
Tuberculosis remains a major public health problem in both developing and developed countries. Drug-resistant tuberculosis is an increasing health problem and serious challenge to tuberculosis (TB) control programs. Information about the susceptibility pattern of Mycobacterium tuberculosis isolates against anti-tuberculosis drugs is an important aspect to TB control. The objectives of the s...
متن کاملRadiological Dependence in Diagnosis of Pulmonary Tuberculosis
Introduction: Diagnosis of tuberculosis under the National Tuberculosis Control Program (NTCP) is based on microscopic examination of sputum. The main objective of this study was to assess the perception of physicians and patients regarding the importance of chest X-ray in diagnosing pulmonary tuberculosis with a view to suggest corrective measures. Materials & Methods: One hundred and eight...
